BON E MARRO W TO DIFFERENTIATE TORV^ARD CARDIOMYOCYTE S IN VITRO
Heart is high differential tissue so that the regenaration df adult cardiumydcytes is limited, specially
affter myocardial infarction. Cell tharepy includes mesenchymal stem cells with their potency has become
an increasing field of interest. Objective: (l)Appling a protocdl use 5 - azacytidine induced mesenchymal
stem cells from human bone marrow to differentiate torward into cardiomyocytes in vitro. (2)lnvestigating
a successful level of this protocol that differentes MSCs toward cardiomyocyte - like cells. Method: In vitro
model. Mesenchymal stem cells from human bone marrow at passeges 2 were induced by 5 - azacytidine
lOpM in 24 hours, were colected to exam affter that about 3 - 4 weeks. Using RT - PCR,
immunohistochemistry (IHC) and transmission electren microscopy (TEM) to appreciate the results of the
differentiation process. Concludes:(1) 5 - azacytidin induced MSCs towards a cardiomyogenic lineage in
vitro. (2) Results of defferentiation process are observated in three myo - cardiomyocyte - specific levels.
The induced cells increated gens expression (MEF2A, 2C, 2D va GATA4); enhanced skeleton cytoplasm
protein expression - Desmin, appeared intermediate filamens in cytoplasm, but not sarcomeres.

tU 2 ng/ml trd xudng (gia tri binh thudng) tang theo thdi gian, cao nhat d 3 thang sau diiu tri (88,8%o) va van
duy tri d mUc cao 6 thang sau diiu tri (88,3%o). Ket luan: nong do SCC - Ag huyet thanh la rat cd gia tri trong
viee theo ddi ket qua dieu tri ung thu cd td eung.
